Mechanism of Action
Abacavir is a carbocyclic guanosine analogue that follows an unusual activation route, being converted by adenosine phosphotransferase and cytosolic deaminase to carbovir monophosphate and then to carbovir triphosphate. Carbovir triphosphate competes with deoxyguanosine triphosphate for incorporation by HIV-1 reverse transcriptase. The carbocyclic ring lacks a 3-hydroxyl group, so incorporation terminates viral DNA chain extension.
