Mechanism of Action
Acyclovir is an acyclic guanosine analogue that is selectively monophosphorylated by herpesvirus-encoded thymidine kinase, so activation is largely confined to infected cells. Host guanylate kinase and other cellular kinases complete conversion to acyclovir triphosphate, which competitively inhibits viral DNA polymerase far more potently than the host enzyme. Incorporation into the elongating DNA strand causes obligate chain termination and inactivates the polymerase.
