Mechanism of Action
Amodiaquine is a 4-aminoquinoline that concentrates in the acidic food vacuole and blocks haem polymerisation, in the same manner as chloroquine. Its principal active metabolite, desethylamodiaquine, sustains the effect with a substantially longer half-life. Structural differences in the side chain preserve useful activity against many chloroquine-resistant Plasmodium falciparum strains.
