Mechanism of Action

Amoxicillin binds penicillin-binding proteins 1a, 1b and 3, inhibiting the transpeptidase reaction that cross-links peptidoglycan chains in the bacterial cell wall. The para-hydroxy group on the phenylglycine side chain extends the spectrum beyond benzylpenicillin to include many Enterobacterales, Haemophilus influenzae and Helicobacter pylori. It is readily hydrolysed by class A beta-lactamases, which is why it is so often co-formulated with clavulanate potassium.
