Mechanism of Action
Amphotericin B binds ergosterol in the fungal membrane and forms ion-permeable channels, and also extracts ergosterol directly out of the bilayer into extramembranous aggregates. Both mechanisms collapse the transmembrane electrochemical gradient and cause rapid fungicidal killing. Residual affinity for mammalian cholesterol underlies its nephrotoxicity, which is what drove development of the liposomal and lipid-complex presentations.
