Mechanism of Action
Artemether contains an endoperoxide bridge that is cleaved by ferrous iron and haem liberated during parasite digestion of host haemoglobin, generating carbon-centred free radicals. These radicals alkylate essential parasite proteins and haem, and disrupt PfATP6-linked calcium homeostasis and redox balance. The result is rapid killing across ring, trophozoite and early gametocyte stages, giving the fastest parasite clearance of any antimalarial class.
