Mechanism of Action

Artemisinin carries a 1,2,4-trioxane endoperoxide bridge that is reductively activated by ferrous iron and haem released during parasite haemoglobin catabolism. The resulting carbon-centred radicals alkylate a broad range of parasite proteins and overwhelm redox defences. Artemisinin itself is now used almost entirely as the synthetic starting material for its more potent derivatives rather than as a finished drug.
