Mechanism of Action
Artesunate is a hemisuccinate prodrug that is rapidly hydrolysed in plasma to dihydroartemisinin, the active endoperoxide. Iron-mediated cleavage of the peroxide bridge generates alkylating radicals that damage parasite membrane proteins, haem detoxification pathways and the sarcoplasmic calcium ATPase. Its water solubility permits intravenous dosing, delivering the fastest achievable parasite biomass reduction in severe falciparum malaria.
