Mechanism of Action
Avibactam is a diazabicyclooctane inhibitor that carbamoylates the active-site serine of beta-lactamases through reversible covalent binding, with slow deacylation that regenerates intact inhibitor rather than destroying it. This recycling behaviour distinguishes it from the suicide-substrate clavams and sulfones. It inhibits Ambler class A, class C and some class D enzymes, including KPC carbapenemases and AmpC, though not metallo-beta-lactamases.
