Mechanism of Action

Bacitracin binds the lipid carrier undecaprenyl pyrophosphate and blocks its dephosphorylation, halting the recycling step that regenerates the carrier needed to ferry peptidoglycan subunits across the membrane. Cell wall synthesis stops and Gram-positive organisms are killed. Zinc complexation stabilises the peptide against degradation and enhances antibacterial potency, which is why the zinc salt rather than the free base is used in nearly all commercial products.
