Mechanism of Action
Benznidazole is activated inside Trypanosoma cruzi by a parasite-specific type I nitroreductase, NTR-1, which reduces the nitro group to reactive glyoxal and dialdehyde metabolites. These electrophiles form covalent adducts with parasite DNA, lipids and proteins, and the parasite lacks the catalase and glutathione peroxidase defences that protect mammalian cells. This selective bioactivation explains both the antiparasitic effect and the resistance seen where NTR-1 activity is lost.
