Mechanism of Action
Caspofungin non-competitively inhibits beta-(1,3)-D-glucan synthase, the enzyme complex that polymerises the principal structural glucan of the fungal cell wall. Loss of beta-(1,3)-D-glucan leaves the wall osmotically unstable, producing fungicidal lysis of Candida species and growth arrest at the hyphal tips of Aspergillus. Because mammalian cells have no cell wall and no homologous enzyme, the class has a wide therapeutic margin and few off-target effects.
