Mechanism of Action

Cefazolin binds penicillin-binding proteins 1 and 3 and inhibits the transpeptidase reaction that cross-links peptidoglycan strands, causing cell wall failure and lysis. Its tetrazolyl and thiadiazolyl substituents deliver high plasma concentrations and a longer half-life than other first-generation parenteral cephalosporins. Activity centres on methicillin-susceptible Staphylococcus aureus, streptococci and community Enterobacterales.
