Mechanism of Action
Ceftaroline fosamil is an N-phosphono prodrug rapidly converted in plasma to active ceftaroline. Unlike earlier cephalosporins, ceftaroline binds the altered penicillin-binding protein PBP2a of methicillin-resistant Staphylococcus aureus by inducing an allosteric conformational change that opens the active site. This gives genuine anti-MRSA activity within the beta-lactam class, alongside conventional PBP binding in streptococci and Gram-negative organisms.
