Mechanism of Action

Chloroquine is a weak base that concentrates several thousand-fold in the acidic digestive vacuole of the malaria parasite. There it binds ferriprotoporphyrin IX and prevents its polymerisation into inert haemozoin, so free haem and chloroquine-haem complexes accumulate and lyse parasite membranes. Resistance in Plasmodium falciparum arises chiefly from mutations in the PfCRT transporter that pump the drug out of the vacuole.
