Mechanism of Action

Ciclopirox is not a sterol-pathway inhibitor; it chelates polyvalent cations, particularly Fe3+ and Al3+, and thereby inactivates metal-dependent fungal enzymes including cytochromes and catalase. Loss of these enzymes disrupts mitochondrial electron transport, membrane transport and peroxide degradation, killing the organism. Because the mechanism is distinct from the azoles and allylamines, there is no cross-resistance, and it retains activity against bacteria and Malassezia as well as dermatophytes.
