Mechanism of Action
Clofazimine is a riminophenazine that binds preferentially to mycobacterial DNA at guanine-rich sequences and inhibits template function. It is also reduced by the type II NADH dehydrogenase of the respiratory chain and then reoxidised by oxygen, generating reactive oxygen species that damage membrane phospholipids. The combination of DNA binding and redox cycling produces slow bactericidal activity against Mycobacterium leprae and Mycobacterium tuberculosis, alongside a marked anti-inflammatory effect.
