Mechanism of Action

Daclatasvir binds domain I of the hepatitis C NS5A protein, a non-enzymatic phosphoprotein essential to assembly of the viral replication complex. Binding disrupts both formation of new replication complexes on the endoplasmic reticulum membrane and the later virion assembly and secretion steps. The dual block on RNA replication and assembly produces very rapid multi-log declines in viral load when combined with a polymerase inhibitor.
