Mechanism of Action
Dalbavancin binds the D-alanyl-D-alanine terminus of lipid II and nascent peptidoglycan, blocking transglycosylation and transpeptidation and halting cell wall assembly. Its lipophilic side chain anchors the molecule in the bacterial membrane, increasing local concentration and target residence time. The same lipophilicity plus high plasma protein binding gives a terminal half-life of roughly two weeks, which is its defining clinical property.
