Mechanism of Action
Dapsone is a sulfone analogue of para-aminobenzoic acid that competitively inhibits dihydropteroate synthase in the bacterial folate biosynthesis pathway. Blocking dihydrofolate formation starves the organism of the tetrahydrofolate needed for purine and thymidine synthesis, giving bacteriostatic activity against Mycobacterium leprae. It also exerts a separate anti-inflammatory effect by suppressing neutrophil myeloperoxidase and chemotaxis, which underpins its dermatological uses.
