Mechanism of Action
Daptomycin inserts its decanoyl lipid tail into the Gram-positive cytoplasmic membrane in a strictly calcium-dependent manner and then oligomerises to form membrane-spanning aggregates. These aggregates cause rapid potassium efflux, membrane depolarisation and arrest of DNA, RNA and protein synthesis, killing the cell without lysis. Pulmonary surfactant sequesters and inactivates the molecule, which is why it has no role in pneumonia.
