Mechanism of Action
Darunavir is a second-generation non-peptidic protease inhibitor whose bis-tetrahydrofuran group forms extensive hydrogen bonds with the backbone atoms of the HIV-1 protease active site. Because these contacts are with conserved main-chain atoms rather than side chains, activity is retained against many multidrug-resistant protease variants. Inhibition prevents gag-pol polyprotein cleavage and produces immature, non-infectious virus particles.
