Mechanism of Action
Delamanid is a nitroimidazooxazole prodrug bioactivated by the mycobacterial deazaflavin-dependent nitroreductase Ddn using cofactor F420. The activated species inhibits synthesis of methoxy-mycolic and keto-mycolic acids, disabling cell wall assembly. Reactive nitrogen intermediates released during activation add respiratory poisoning activity against non-replicating bacilli.
