Mechanism of Action

Demeclocycline binds the 30S ribosomal subunit and blocks aminoacyl-tRNA association with the A site, giving bacteriostatic inhibition of protein synthesis. Independently of its antibacterial action it antagonises vasopressin activity at the renal collecting duct, which is the basis of its non-antibacterial nephrology use. The 7-chloro, 6-demethyl structure gives a longer half-life than tetracycline.
