Mechanism of Action

Dicloxacillin carries a dichlorophenyl isoxazolyl side chain that provides even greater steric protection against staphylococcal beta-lactamase than cloxacillin. The protected beta-lactam ring acylates penicillin-binding proteins and prevents the transpeptidation step of peptidoglycan assembly. High plasma protein binding and superior oral bioavailability relative to cloxacillin make it the preferred oral agent in this subclass.
