Mechanism of Action
Dolutegravir binds the active site of HIV-1 integrase and chelates the two magnesium ions held by the catalytic DDE triad within the intasome. This blocks the strand transfer step in which viral DNA ends are joined to host chromosomal DNA, halting integration and productive infection. Its slow dissociation from the intasome underpins a high barrier to resistance relative to earlier integrase inhibitors.
