Mechanism of Action

Emtricitabine is the 5-fluorinated analogue of lamivudine and is phosphorylated by cellular kinases to emtricitabine 5-triphosphate. The triphosphate competes with deoxycytidine triphosphate for incorporation into nascent viral DNA by HIV-1 reverse transcriptase and hepatitis B polymerase. Absence of a 3-hydroxyl on the oxathiolane ring makes incorporation an obligate chain-terminating event.
