Mechanism of Action

The fluorinated and chlorinated phenyl isoxazolyl side chain resists hydrolysis by staphylococcal penicillinase while retaining affinity for penicillin-binding proteins. Acylation of PBP transpeptidase domains halts peptidoglycan cross-linking and triggers autolytic cell death. The fluorine substitution improves oral absorption and tissue penetration compared with cloxacillin.
