Mechanism of Action

Griseofulvin binds fungal tubulin and associated microtubule-associated proteins, disrupting mitotic spindle assembly and arresting the dermatophyte cell cycle in metaphase. It also interferes with microtubule-dependent transport of cell wall precursors, producing distorted, stunted hyphae. Because it deposits preferentially in newly formed keratin, activity is confined to keratinised tissue and to dermatophytes only.
