Mechanism of Action
Josamycin is a 16-membered macrolide that binds the 50S ribosomal subunit and blocks the peptidyl transferase centre, halting nascent chain elongation. Its 16-membered ring means it is not inactivated by the inducible erm-mediated MLSb resistance seen with 14-membered macrolides in some staphylococcal strains. Activity is bacteriostatic against Gram-positive cocci, Mycoplasma and Chlamydia.
