Mechanism of Action

Ledipasvir is a bridged bis-imidazole NS5A inhibitor that binds domain I of the hepatitis C NS5A protein near the dimer interface. This prevents assembly of functional membrane-associated replication complexes and also impairs virion assembly and release. Potency is highest against genotypes 1, 4, 5 and 6, and it is used only in combination with sofosbuvir to protect against rapid resistance selection.
