Mechanism of Action
Lopinavir is a peptidomimetic that binds the C2-symmetric active site of HIV-1 protease and blocks hydrolysis of gag and gag-pol polyprotein precursors. The resulting virions retain uncleaved structural and enzymatic proteins and are non-infectious. Lopinavir is extensively metabolised by CYP3A4, which is why it is invariably co-formulated with low-dose ritonavir as a pharmacokinetic booster.
