Mechanism of Action

Luliconazole inhibits fungal lanosterol 14-alpha-demethylase and thereby ergosterol biosynthesis, but its R-enantiomer binds with exceptional potency, giving minimum inhibitory concentrations against dermatophytes an order of magnitude below older imidazoles. The ketene dithioacetal moiety appears to contribute additional membrane-level activity. Its high keratin affinity produces prolonged retention in the stratum corneum, allowing short once-daily courses.
