Mechanism of Action

Lumefantrine is thought to interfere with the polymerisation of haem into inert haemozoin inside the parasite food vacuole, allowing cytotoxic free haem and reactive oxygen species to accumulate. Its long elimination half-life clears residual parasitaemia after the short-acting artemisinin partner has collapsed parasite biomass. This partner-drug tail is what protects the artemisinin component from resistance selection.
