Mechanism of Action

Mefloquine is a 4-quinoline methanol that concentrates in the parasite food vacuole and is believed to disrupt haem detoxification and to interfere with parasite endocytosis of host cytosol. It also inhibits the 80S ribosome, contributing a second point of attack. Its long elimination half-life makes it an effective partner drug and a once-weekly prophylactic option where the resistance picture allows.
