Mechanism of Action

Mezlocillin is an acylureido penicillin whose imidazolidinyl-urea side chain enhances penetration of the Gram-negative outer membrane and affinity for penicillin-binding protein 3. Inhibition of PBP3 blocks septal peptidoglycan synthesis, producing filamentous cells that subsequently lyse. Its spectrum covers many Enterobacterales, Pseudomonas aeruginosa and Bacteroides species, although it remains susceptible to most beta-lactamases.
