Mechanism of Action

Minocycline binds reversibly to the 30S ribosomal subunit and blocks attachment of aminoacyl-tRNA to the ribosomal A site, halting peptide chain elongation. Its 7-dimethylamino group and absence of a C6 hydroxyl give the highest lipophilicity of the classical tetracyclines and correspondingly strong tissue and CNS penetration. Separate anti-inflammatory and matrix metalloproteinase-inhibiting effects underpin its dermatological and periodontal use.
