Mechanism of Action

Nalidixic acid inhibits the A subunit of bacterial DNA gyrase, blocking the religation step of DNA supercoiling and generating lethal double-strand breaks. As the original naphthyridone quinolone it lacks a fluorine at C6, which limits potency and tissue penetration relative to fluorinated successors. Activity is essentially confined to Gram-negative urinary pathogens.
