Mechanism of Action
Neomycin binds irreversibly to the 16S rRNA of the 30S ribosomal subunit, causing misreading of the mRNA codon and incorporation of incorrect amino acids into nascent proteins. Aberrant proteins insert into the cell membrane, increasing permeability and driving further aminoglycoside uptake in a self-amplifying lethal cycle. Killing is concentration dependent and bactericidal.
