Mechanism of Action

Netilmicin binds the 30S ribosomal subunit and induces codon misreading and premature termination of translation, generating defective membrane proteins and lethal permeability changes. The 1-N-ethyl substitution on sisomicin sterically hinders several aminoglycoside-modifying enzymes, preserving activity against some gentamicin-resistant Gram-negative isolates. Bactericidal action is concentration dependent with a prolonged post-antibiotic effect.
