Nevirapine molecular structure

Nevirapine

Also known as: Viramune, Nevimune, Nevipan (component of Duovir-N, Triomune)

CAS Number: 129618-40-2

Non-Nucleoside Reverse Transcriptase Inhibitor (NNRTI)

Pharmacopeia Standards

USPIPEPBPWHO

Mechanism of Action

Nevirapine is a dipyridodiazepinone that binds directly to the non-nucleoside allosteric pocket of HIV-1 reverse transcriptase about ten angstroms from the polymerase active site. The resulting conformational distortion of the catalytic aspartate triad blocks RNA-dependent and DNA-dependent DNA polymerase activity without competing with nucleoside triphosphates. Activity is restricted to HIV-1, and single point mutations such as K103N or Y181C confer high-level resistance.
