Mechanism of Action

Nitroxoline is an 8-hydroxyquinoline that chelates divalent cations, principally zinc, magnesium and iron, stripping the metal cofactors that bacteria need for metalloenzyme function and for biofilm matrix stability. This metal-chelating action inhibits RNA polymerase activity and prevents adhesion and biofilm formation on urothelium and catheter surfaces, which distinguishes it from conventional urinary antibiotics. It reaches high concentrations in urine while producing little systemic exposure.
