Mechanism of Action
Norfloxacin blocks bacterial DNA gyrase, the type II topoisomerase responsible for introducing negative supercoils ahead of the replication fork. Stabilisation of the cleaved DNA-gyrase complex converts the enzyme into a chromosome-fragmenting poison. Urinary concentration greatly exceeds serum levels, which shaped its historical clinical positioning.
