Mechanism of Action

Oxacillin was the first isoxazolyl penicillin, using an unsubstituted phenyl isoxazolyl side chain to block access of staphylococcal penicillinase to the beta-lactam carbonyl. The surviving ring acylates penicillin-binding proteins and arrests peptidoglycan transpeptidation. Susceptibility to oxacillin is the defining laboratory criterion separating methicillin-susceptible from methicillin-resistant Staphylococcus aureus.
