Mechanism of Action

Procaine benzylpenicillin is a sparingly soluble 1:1 salt that dissolves slowly from the intramuscular depot, releasing benzylpenicillin over roughly twenty-four hours. Liberated benzylpenicillin inactivates penicillin-binding proteins and prevents peptidoglycan cross-linking. The procaine moiety also provides local anaesthesia at the injection site, improving tolerability of the large-volume suspension.
