Mechanism of Action

Remdesivir is a phosphoramidate ProTide prodrug of an adenosine C-nucleoside analogue, converted intracellularly to the active triphosphate GS-443902. The triphosphate is incorporated by coronavirus RNA-dependent RNA polymerase in place of adenosine triphosphate and causes delayed chain termination three nucleotides downstream. The 1-cyano substituent also creates a steric clash that resists removal by the viral proofreading exonuclease.
