Rifabutin molecular structure

Rifabutin

Also known as: Mycobutin, Rifabin, R-Butin, Ricobutin

CAS Number: 72559-06-9

Antitubercular / Antimycobacterial (Rifamycin, Ansamycin)

Pharmacopeia Standards

USPEPIP

Mechanism of Action

Rifabutin binds the beta subunit of bacterial DNA-dependent RNA polymerase and blocks the elongation of nascent messenger RNA, shutting down transcription. Its spiro-piperidyl ansamycin structure retains activity against many rifampicin-resistant strains carrying certain rpoB mutations. Rifabutin is a substantially weaker inducer of cytochrome P450 3A4 than rifampicin, which is why it is preferred where antiretroviral or immunosuppressant co-administration is a concern.
