Ritonavir molecular structure

Ritonavir

Also known as: Norvir, Ritomune (component of Kaletra, Aluvia, Paxlovid, boosted PI regimens)

CAS Number: 155213-67-5

HIV Protease Inhibitor / Pharmacokinetic Enhancer

Pharmacopeia Standards

USPIPEPBPWHO

Mechanism of Action

Ritonavir is a peptidomimetic inhibitor that binds the catalytic site of HIV-1 aspartyl protease, preventing cleavage of the gag and gag-pol polyproteins and yielding immature, non-infectious virions. At the low doses used clinically its dominant role is pharmacokinetic boosting through potent mechanism-based inhibition of cytochrome P450 3A4 and inhibition of P-glycoprotein. This raises plasma exposure of co-administered protease inhibitors and other CYP3A4 substrates.
