Mechanism of Action

Spectinomycin binds helix 34 of the 16S rRNA in the 30S subunit and blocks the EF-G-catalysed translocation step of protein synthesis without causing the codon misreading typical of true aminoglycosides. Inhibition is bacteriostatic and reversible, which distinguishes it pharmacologically from gentamicin-class agents. It lacks the nephrotoxic and ototoxic profile associated with aminoglycosides proper.
