Mechanism of Action
Sulfadoxine is a structural analogue of para-aminobenzoic acid that competitively inhibits parasite dihydropteroate synthase, blocking folate biosynthesis at an early step. Because malaria parasites must synthesise folate de novo rather than salvage it, this starves them of the cofactors needed for DNA synthesis. Paired with pyrimethamine, which inhibits the downstream dihydrofolate reductase, the sequential blockade is markedly synergistic.
